Enzyme activity was measured in a buffer containing 100 mM Sodium acetate (pH 5.5), 2 mM EDTA, 8 mM DTT, 0.05% Brij, and 20 μM substrate (Z-FRAMC), with 3.75-30 ng/μl Cathepsin F. Reaction was carried out at 30°C for 40-120 min, and fluorescence signal was measured at Ex360 nm/Em460 nm. Enzyme was preactivated in 50 mM sodium acetate pH 4.5, 5 mM DTT buffer for 90 min at RT prior to reaction.,

Cathepsin F also known as CTSF is a cysteine protease enzyme with a molecular mass of approximately 44 kDa. It belongs to the papain-like protease family and exhibits proteolytic activity. This protein localizes to the lysosomes where it participates in protein degradation. Cathepsin F shows expression in various tissues including liver kidney and spleen suggesting a broad role in protein catabolism.

Cathepsin F functions in protein breakdown processes within the lysosomal compartment. It actively participates in the formation and maturation of the autophagosomes.
