The Hairy protein also known as HER or Hairy/enhancer of split-related protein is a basic helix-loop-helix (bHLH) transcription factor with a molecular mass of approximately 30 kDa. This protein primarily functions by binding to specific DNA sequences thereby repressing the transcription of target genes. It plays a significant role in segmentation during embryonic development. Hairy is expressed prominently in various tissues including those involved in neurogenesis and mesodermal segmentation.
Biological function summary
Hairy helps regulate the timing of development processes in embryogenesis. It acts as a transcriptional repressor and is part of the Hes family proteins often forming a complex with other bHLH proteins. Through its repressive actions Hairy controls cell fate decisions and maintains the balance between proliferation and differentiation in developing tissues. It regulates gene expression patterns that are essential for precise body plan formation during embryonic development.
Pathways
Hairy is intimately connected through the Notch signaling pathway which is critical for cell fate determination and pattern formation in multicellular organisms. The interaction with Notch signaling components like the Notch receptor and its ligands influences cellular communication necessary for tissue organization. Additionally Hairy is involved in the segmentation clock pathway interacting with proteins such as the Drosophila homolog of the mammalian Hes1 and Hes5 to coordinate segmentation during development.
Hairy has links to genetic conditions such as spondylocostal dysostosis characterized by abnormal spinal segmentation. Mutations affecting the Hairy gene impact proper development by disrupting the balance of signaling pathways which can lead to vertebral anomalies. Its interaction with other Hes family proteins compounds these effects making Hairy a focal point in studying congenital vertebral disorders.
