HELB also known as DNA helicase B plays an important role in DNA replication and repair. This protein has a mass of approximately 112 kDa. It acts as a DNA helicase unwinding the duplex DNA during replication and repair. HELB is expressed mainly in the nucleus of proliferating cells particularly in tissues with high cell division rates like the testes and bone marrow. Important for maintaining genome stability HELB interacts with various DNA structures to facilitate repair processes.
Biological function summary
DNA helicase B takes part in the maintenance of genomic integrity through its helicase activity. HELB does not form a known complex but acts synergistically with other proteins during DNA synthesis and repair. It interacts with replication protein A (RPA) enhancing its ability to assist in DNA repair. The protein influences DNA damage response preventing replication fork stalling and promoting cell cycle progression.
Pathways
HELB functions in important DNA repair pathways like homologous recombination and base excision repair. Through these pathways it collaborates with key proteins such as ATM and ATR kinases. HELB regulates the DNA damage response by modulating the action of these kinases which are vital for cell cycle checkpoints and repair mechanisms.
HELB has associations with cancer and genomic instability disorders. Alterations in HELB expression can impact DNA repair proficiency potentially leading to oncogenesis. Mutations or dysfunctions in HELB may contribute to the development of cancer by allowing DNA damage accumulation. Additionally HELB's interaction with proteins such as BRCA1 indicates its involvement in breast cancer where ineffective repair of DNA damage can promote tumorigenesis.
