Sec8 also known as Exocyst complex component 4 (EXOC4) is a protein with a mass of approximately 100 kDa. It acts as a critical component of the exocyst complex which is essential for the tethering of vesicles to specific sites on the plasma membrane. Sec8 shows high expression in a variety of human tissues including the brain kidney and intestine. By anchoring vesicles it plays an important role in exocytosis and the regulation of membrane dynamics.
Biological function summary
The Sec8 protein contributes to cellular events by serving as a part of the exocyst complex a multiprotein assembly involved in vesicle trafficking. This complex consists of eight proteins including Sec3 Exo70 and Sec15. The assembly coordinates precise vesicle delivery facilitating processes such as cell growth migration and polarity establishment. Its activity is important for the proper functioning of secretory pathways ensuring communication and material transport across cell membranes.
Pathways
Sec8 operates within the secretory pathway that manages delivery and distribution of materials inside cells. It is also involved in the sustainable function of the polarized secretion pathway an essential process that ensures cellular asymmetry and directional growth. Sec8's interaction with proteins like Rab family GTPases is significant in these pathways which are responsible for guiding vesicle movement and anchoring to the target membrane.
Sec8 associations have been identified in relation to cancer and impaired cognitive function. Alterations in the expression or function of Sec8 may disrupt vesicle trafficking leading to tumorigenesis or neurodevelopmental disorders. For example within cancer this disruption can affect cell adhesion and migration promoting metastasis. Additionally interactions with proteins such as Sec6 another exocyst complex member further illustrate the interconnected impact of Sec8 dysregulation on diseases.
