The Early B-cell Factor 3 (EBF3) also known as COE3 (Collier/Olf/EBF) acts as a transcription factor important for B-cell development and neuronal differentiation. EBF3 with a mass of approximately 63 kDa is expressed in various tissues including the brain and bone marrow. It plays a role in DNA binding through its helix-loop-helix domain and is involved in the regulation of gene expression by interacting with specific promoter regions.
Biological function summary
EBF3 influences several critical cellular processes including neurogenesis and immune function. It contributes to the regulation of neuronal gene expression ensuring proper neuronal identity and function. EBF3 is part of a transcriptional complex with other EBF family members which collectively coordinate the expression of target genes essential for normal cellular differentiation and function.
Pathways
EBF3 integrates into the neurogenesis and immune response pathways. In the neurogenesis pathway it interacts with proteins such as NeuroD1 and Pax6 to influence the fate and differentiation of neural progenitors. Within the immune system EBF3 works alongside related proteins like PAX5 to regulate the development and function of B-cells maintaining immune homeostasis.
EBF3 has links to neurodevelopmental disorders and certain types of leukemia. Mutations or dysregulation of EBF3 can contribute to disorders such as hypotonia and ataxia linked to EBF3-related neurodevelopmental disorder (E3ND). In leukemia EBF3 can cooperate with abnormal signaling pathways to alter B-cell proliferation. In this context it may have interactions with proteins like RUNX1 influencing disease progression and therapeutic outcomes.
