Hepatitis C Virus NS4 also known as nonstructural protein 4 is a component of the Hepatitis C virus important for viral replication and assembly. This protein expressed in infected hepatocytes has an approximate mass of 27 kDa. NS4 plays an integral role in the virus's life cycle impacting processes essential for the formation of viral replication complexes.
Biological function summary
NS4 functions to modulate interactions required for viral polyprotein processing and pathogenicity. It forms a part of the larger nonstructural protein complex interacting with other viral proteins such as NS3 and NS5A. These interactions significantly contribute to membrane rearrangement and the stabilization of the replication complex. The interplay of NS4 with various host cell factors aids in creating a favorable environment for viral replication.
Pathways
NS4 is deeply involved in the viral replication pathway and host immune response modulation. In the viral replication pathway NS4 acts in concert with NS3 which is a serine protease. Additionally it manipulates the host immune mechanisms to evade detection impacting interferon signaling pathways. Through these pathways NS4 supports the continuity of viral propagation within host cells.
NS4 correlates predominantly with Hepatitis C infection which can progress to chronic Hepatitis C and lead to liver cirrhosis or hepatocellular carcinoma. The interaction of NS4 with other viral proteins like NS5A further exacerbates the pathogenic effects of the virus promoting disease progression. Understanding NS4's role aids in the development of effective antiviral therapies targeting these interactions.
