Ten-1 also known as Tensin-1 is a cytoskeletal protein with a mass of around 220 kDa. This protein mainly localizes to focal adhesions structures that connect the extracellular matrix to the cytoskeleton. Ten-1 binds actin filaments and it contains several domains such as a SH2 domain enabling it to interact with phosphotyrosine residues. This protein is expressed in various tissues with higher levels in kidney liver and heart where it plays a part in maintaining cellular architecture and signal transduction.
Biological function summary
Ten-1 functions in cell adhesion and migration by linking the cytoskeleton to integrins at focal adhesions. Sometimes Ten-1 forms complexes with other cytoskeletal proteins helping stabilize cell-extracellular matrix interactions. These complexes are essential for mechanical signal transduction and cellular responses to external stimuli. Expression and regulation of Ten-1 influence cell motility and structural integrity playing roles in tissue morphogenesis and wound healing.
Pathways
Ten-1 significantly impacts the Rho signaling pathway influencing cell shape and motility. It does this through interactions with proteins such as Ras-related C3 botulinum toxin substrate 1 (Rac1) and Rho GTPases modulating actin filament dynamics. Another relevant pathway is the focal adhesion kinase (FAK) pathway where Ten-1 interacts with FAK affecting signaling cascades that regulate cell movement and survival. These pathways show how Ten-1 participates in processes important for cellular adaptation to different environmental conditions.
Alterations in Ten-1 expression or mutations can contribute to cancer progression and metastasis by affecting cell adhesion and migration. Studies indicate that lower levels of Ten-1 correlate with aggressive tumor phenotypes in breast and prostate cancers. Besides cancer Ten-1 interacts with proteins like vinculin which influences its role in tension transmission and force generation in cells potentially contributing to developmental disorders related to focal adhesion dysfunction. Understanding Ten-1's function in these contexts can offer insights into therapies targeting pathological cell migration and adhesion.
