ORMDL3 also known as ORM1-like protein 3 is a transmembrane protein located in the endoplasmic reticulum. The molecular mass of ORMDL3 is approximately 17 kDa. This protein is expressed ubiquitously meaning it is found in most tissues throughout the body. ORMDL3 plays an important role in regulating sphingolipid biosynthesis by inhibiting the serine palmitoyltransferase (SPT) complex which is the first step in the sphingolipid de novo pathway.
Biological function summary
ORMDL3 modulates lipid metabolism and immune response regulation. It does this by being part of a regulatory complex with the serine palmitoyltransferase therefore influencing the production of sphingolipids which are essential components of cell membranes and contribute to cell signaling. Alterations in sphingolipid levels can affect membrane integrity and cell communication impacting cellular function and health.
Pathways
ORMDL3 influences sphingolipid metabolic processes and endoplasmic reticulum stress responses. In the sphingolipid metabolism pathway it directly interacts with SPTLC1 and SPTLC2 core components of the serine palmitoyltransferase enzyme complex. Additionally ORMDL3 indirectly affects the unfolded protein response pathway by influencing levels of sphingolipids which play a role in stress signaling in the endoplasmic reticulum.
Studies link ORMDL3 to asthma and inflammatory bowel disease (IBD). ORMDL3's role in sphingolipid regulation affects inflammation and immune responses processes that are critical in the pathology of these diseases. For example genetic variations in ORMDL3 correlate with increased risk of asthma potentially through interactions with other immune-related proteins such as GSDMB. Changes in ORMDL3 expression levels can influence the severity and progression of inflammatory disorders by modulating the sphingolipid-mediated signaling pathways.
