ZNF423 also known as OAZ or EBF-associated zinc finger protein acts mechanically as a transcription factor that interacts with DNA to regulate gene expression. This protein comprises of multiple zinc finger motifs enabling it to bind specifically to DNA sequences. ZNF423 has a mass of approximately 150 kDa. It is expressed in various tissues including brain and kidney where it plays a vital role in development and functioning of these organs.
Biological function summary
ZNF423 functions as an essential modulator of neurodevelopmental processes. It is a component of the nuclear receptor coactivator complex where it interacts with several other transcription factors to influence neuronal differentiation and proliferation. It exerts its biological function by regulating target genes involved in growth and development. This modulation activity makes ZNF423 important for proper embryonic development.
Pathways
Proteins within the DNA repair and neurotrophin signaling pathways engage with the actions of ZNF423. In the DNA repair pathway ZNF423 interacts with PARP-1 a critical protein in repairing DNA single-strand breaks to maintain genomic integrity. In the neurotrophin signaling pathway it works with proteins such as EBF1 to influence neuronal growth and survival highlighting its role in brain development and function.
Malfunctions in ZNF423 are associated with certain neurodevelopmental disorders and cancers. Nonsense mutations in the ZNF423 gene can lead to nephronophthisis a kidney disorder characterized by inflammation and scarring. Additionally abnormalities in ZNF423 expression often correlate with pediatric high-grade gliomas a form of aggressive brain tumors. In these contexts ZNF423 connects with proteins like SMAD4 impacting tumorigenesis and disease progression.
