RGC-32 also known as response gene to complement 32 is a protein with a molecular weight of approximately 16 kDa. It acts as a cell cycle regulator and transcription coactivator. The protein influences cell division and apoptosis by interacting with other molecules in the cell. RGC-32 is strongly expressed in various tissues including the heart brain liver and kidneys. Its presence in many tissues suggests a widespread role in cellular processes beyond mere response to complement stimuli.
Biological function summary
RGC-32 contributes to cell cycle progression and differentiation. It does not act alone but collaborates with other proteins and enzymes to enable these processes. RGC-32 forms part of a complex that governs key steps in cell cycle regulation influencing cellular proliferation and differentiation pathways. Its involvement in these signaling cascades makes it essential for maintaining proper cell cycle control impacting cellular repair and growth.
Pathways
RGC-32 participates in the regulation of the TGF-β signaling pathway and the Wnt/β-catenin pathway. These two pathways are important for cellular growth differentiation and immune response. Within the TGF-β signaling pathway RGC-32 enhances the pathway's ability to mediate cell cycle arrest and fibrosis. In connection with the Wnt/β-catenin pathway RGC-32 modulates transcriptional activity that affects cell proliferation. The protein interacts with Smad proteins in TGF-β signaling forming part of the network that responds to growth factors.
RGC-32 is associated with conditions such as cancer and fibrosis. In cancer its overexpression or dysregulation may lead to uncontrolled cell proliferation and tumor development often through interactions with oncogenes and tumor suppressors. In fibrosis RGC-32 contributes to excessive extracellular matrix production playing a part in tissue scarring processes. The protein's interactions with TGF-β signaling are critical in fibrotic disorders while its connection with cyclin-dependent kinases links it potentially to oncogenesis.
