TMEM63A also known as Transmembrane protein 63A is a mechanosensitive ion channel. It is part of the TMEM family and has an approximate mass of around 85 kDa. This protein locates mainly in the plasma membrane where it plays a role in regulating ion influx in response to mechanical stimuli. TMEM63A expresses highly in tissues such as the kidney and brain indicating its significance in these areas. It allows the passage of cations contributing to maintaining cellular homeostasis.
Biological function summary
TMEM63A helps sense and respond to mechanical changes in the environment working as part of a complex with other ion channels. This function is vital for cellular processes that require reaction to mechanical stress like cell migration and adhesion. In the kidney TMEM63A contributes to maintaining the filtration barrier's integrity which is essential for proper kidney function. In the nervous system its mechanosensitivity may play a role in neural signal regulation.
Pathways
TMEM63A involves itself in mechanotransduction pathways by participating in converting mechanical signals into biochemical signals. It engages with pathways related to cellular response to mechanical stimuli and osmotic balance. It interacts with other proteins such as PIEZO1 within these pathways to modulate cellular response to changes in the mechanical environment. Another associated protein is TRPV4 which also participates in cellular osmoregulation and mechanosensation.
TMEM63A associates with conditions involving disrupted mechanosensation or ion balance. Mutations in TMEM63A link to hereditary kidney disorders like nephrotic syndrome where it may cooperate with proteins involved in podocyte structure such as nephrin. Additionally abnormalities in TMEM63A are under investigation in some neurodevelopmental disorders suggesting interactions with proteins like SHANK3 an important player in synaptic development and function.
