IRG1 also known as Immune-Responsive Gene 1 protein has a molecular weight of approximately 50 kDa. This protein is expressed largely in immune cells such as macrophages and dendritic cells. Mechanically IRG1 catalyzes the production of itaconate from cis-aconitate during the citric acid cycle an action that has gained significant interest due to its immunomodulatory effects. The localization of IRG1 in immune cells suggests a role in the response to pathogenic threats and regulation of inflammation.
Biological function summary
IRG1 is important for immune response regulation by producing itaconate an anti-inflammatory metabolite. Itaconate acts on innate immune signaling pathways to dampen excessive inflammatory responses. Though IRG1 is not part of a larger protein complex it interacts closely with cellular metabolism. This interaction plays a significant role in metabolic reprogramming during infections thereby influencing cytokine production and the activation state of cells.
Pathways
IRG1 plays a significant role in the tricarboxylic acid (TCA) cycle and is associated with the innate immune response. The protein shifts cellular metabolism towards cataplerosis to accommodate the need for itaconate production. IRG1 is connected to regulatory proteins such as the transcription factor NRF2 which mediates antioxidative responses. Itaconate a product of IRG1 activity can modulate pathways like the NF-kB signaling pathway attenuating inflammatory responses.
IRG1 has been implicated in atherosclerosis and tuberculosis. In atherosclerosis IRG1 contributes to the anti-inflammatory state within macrophages which can alter plaque formation. Through itaconate IRG1 interacts with inflammasome components potentially impacting the pathology of the disease. In tuberculosis IRG1-induced itaconate production helps in macrophage defense against Mycobacterium tuberculosis. Additionally pathways involving IRG1 often intersect with proteins like HIF-1α under hypoxic conditions relevant to the progression and management of these diseases.
