Tollip also known as Toll-interacting protein acts as an important regulator in the immune response. This protein weighs approximately 30 kDa and is found expressed in several tissues especially in the lung liver and spleen. Tollip binds to various ubiquitinated proteins and plays a role in the endocytosis process assisting in protein trafficking. It functions by modulating the signaling pathways that involve innate immunity primarily by interacting with Toll-like receptors (TLRs).
Biological function summary
Toll-interacting protein participates in the negative regulation of the immune response preventing overactivation that could lead to inflammation. Tollip forms a complex with TLRs and IL-1 receptor pathway proteins to control inflammatory signaling. Through its action Tollip binds to Tom1 another protein that links Tollip to phosphatidylinositol-3-phosphate. By regulating these pathways Tollip assists in maintaining a balanced immune response.
Pathways
The protein engages in the TLR signaling pathway and the IL-1 receptor pathway. Both pathways are key in initiating immune responses against pathogens. Tollip as part of these pathways directly associates with proteins like IRAK-1 where it modulates the activity and ensures controlled signaling. This regulation prevents excessive or unnecessary activation that can result in immune-related damage.
Tollip links to conditions such as inflammatory bowel disease and sepsis. Both conditions involve significant immune dysregulation. In these disease states Tollip interacts with proteins like MyD88 affecting the response to pathogens and inflammatory stimuli.
