The Secretin receptor (SCTR) also known as the secretin R is a G protein-coupled receptor (GPCR) with a typical seven-transmembrane domain structure. It weighs around 52 kDa and primarily facilitates the action of secretin a hormone involved in various physiological functions. The receptor is expressed in multiple body tissues such as the pancreas stomach and certain regions of the brain. This distribution allows it to mediate the diverse roles of secretin as part of both local and systemic responses.
Biological function summary
The Secretin receptor plays an integral role in regulating fluid and electrolyte balance as well as modulating gastric acid secretion. It does not form part of a complex but operates in concert with other receptors that shape digestive processes. When secretin binds to SCTR it initiates cellular responses that influence pancreatic bicarbonate secretion which is essential for neutralizing stomach acid and creating an optimal environment for intestinal enzymes.
Pathways
The Secretin receptor is chiefly involved in the cAMP signaling pathway. This pathway engages adenylyl cyclase and elevates cAMP levels leading to protein kinase A (PKA) activation. SCTR works alongside other receptors such as the vasoactive intestinal peptide receptor (VIPR) to regulate these processes. This coordinated interaction drives cellular outcomes like enzyme secretion and motility within gastrointestinal tissues.
Alterations in SCTR function have been linked to pancreatic adenocarcinoma and congenital diarrhea syndromes. In these conditions receptor activity may affect signaling pathways and disrupt normal physiological balances. Research indicates that other proteins such as CFTR (cystic fibrosis transmembrane conductance regulator) align with SCTR function particularly in diseases involving disrupted fluid secretion and ion transport. Understanding these connections can help in developing targeted therapies and enhancing our grasp of gastrointestinal disorders.
