ARH3 also known as ADP-ribosylhydrolase 3 is an enzyme involved in the removal of ADP-ribose groups from proteins a process known as de-ADP-ribosylation. It has a mass of approximately 39 kDa. The enzyme shows widespread expression in human tissues with higher levels in the brain heart and skeletal muscles. This expression pattern indicates its important role in various cellular processes across different tissue types.
Biological function summary
ARH3 catalyzes the hydrolysis of ADP-ribose units attached to serine residues on target proteins reversing the actions of PARPs (poly ADP-ribose polymerases) especially PARP1. By doing this it plays a role in regulating the post-translational modification of proteins. Although not considered a part of a permanent complex ARH3 often interacts with other protein factors involved in DNA repair and cellular stress responses suggesting its dynamic involvement in cellular regulation.
Pathways
This enzyme functions in DNA repair and cellular stress response pathways. ARH3's activity complements the work of PARP1 by removing ADP-ribose groups added during the DNA damage response. This enzymatic balance is essential to prevent excessive ADP-ribosylation that can affect DNA repair processes. The interaction between ARH3 and PARP1 illustrates the coordinated efforts required to maintain genomic stability and respond to cellular stress.
Mutations or dysregulation of ARH3 are linked to neurodegenerative disorders and cancer. In neurological diseases proteins modified by ARH3-driven de-ADP-ribosylation may alter neuronal survival and function. Similarly in cancer improper ARH3 activity could lead to uncontrolled cell proliferation due to a faulty DNA damage response. Tumor protein 53 (TP53) might interact with ARH3 indicating potential pathways affected in cancerous conditions.
