HIV-2 Nef also known as Negative Factor is a protein encoded by the HIV-2 virus. It has a molecular mass of approximately 27 kDa. The protein expresses in infected host cells particularly in T lymphocytes and macrophages which are critical components of the immune system. HIV-2 Nef plays an important role in the regulation of viral replication and the downregulation of cell surface molecules such as CD4 and major histocompatibility complex class I (MHC-I).
Biological function summary
HIV-2 Nef is a multifunctional protein that impacts several cellular signaling pathways. It is known to interact with other viral and cellular proteins often forming complexes that modulate signaling pathways to promote viral survival and replication. Nef assists in manipulating the host cell environment which benefits the viral life cycle by preventing apoptosis and enhancing viral egress.
Pathways
HIV-2 Nef engages in critical cellular pathways by interacting with proteins like Src family kinases and PAK2. These interactions alter the actin cytoskeleton and enhance viral spread. It influences pathways such as the MAPK/ERK pathway affecting cell activation proliferation and survival. These pathways are essential for sustaining the viral infection and supporting the destruction of immune cells.
HIV-2 Nef plays a significant role in the pathogenesis of HIV-2 infection and acquired immunodeficiency syndrome (AIDS). It contributes to immune system evasion allowing the virus to persist in the host. Nef's modulation of proteins like CD4 and MHC-I disrupts immune recognition helping the virus avoid immune surveillance. Additionally its interaction with cellular components like Src family kinases links Nef to progression and severity of HIV-related disorders.
