The heterogeneous nuclear ribonucleoprotein M (HNRNPM) is a multifunctional RNA-binding protein also known as glycoprotein p37. It belongs to the hnRNP family. HNRNPM has a molecular mass of approximately 72 kDa. This protein is ubiquitously expressed in various tissues with higher levels in organs like the brain kidney and liver. HNRNPM localizes mainly in the nucleus but shuttles between the nucleus and cytoplasm depending on its phosphorylation state and the cellular context.
Biological function summary
HnRNP M plays key roles in RNA splicing RNA stability and the transport of mRNA molecules from the nucleus to the cytoplasm. It acts as part of the spliceosome complex and interacts with pre-mRNA to regulate alternative splicing which is essential for generating protein diversity. HNRNPM influences the exon inclusion or exclusion in the pre-mRNA affecting the formation of mature mRNA. This regulation capability connects it with processes such as gene expression and cellular response to external signals.
Pathways
HNRNPM has significant roles in RNA splicing and processing pathways. Its involvement in alternative splicing links it to the regulation of the MAPK/ERK signaling pathway. In these pathways HNRNPM coordinates with other proteins like SR proteins and other hnRNP family members ensuring efficient mRNA maturation and transport. These activities highlight its importance in maintaining proper cellular functioning and responding to signaling events.
Research connects HNRNPM to cancer and neurodegenerative diseases. Abnormal expression or mutations in HNRNPM associate with tumor progression metastasis and poor prognosis in various cancers. It often interacts with proteins like E-cadherin and beta-catenin impacting cell adhesion and migration. Additionally dysregulation of HNRNPM expression might contribute to the pathogenesis of neurodegenerative conditions like Alzheimer's disease potentially linking it to proteins such as amyloid precursor protein (APP) through altered RNA processing activities.
