SPOCK1 also known as Testican-1 is a proteoglycan involved mechanically in the regulation of cell-matrix interactions. It's a protein of approximately 54 kDa mass. SPOCK1 is expressed in various tissues with significant expression in the brain and skeletal muscle. In the extracellular matrix it forms part of the calcium-binding protein family having an active role in modulating matrix metalloproteinases (MMPs) and serine protease activities.
Biological function summary
SPOCK1 influences cellular adhesion migration and communication. It interacts with several extracellular matrix components participating actively in the structural organization of tissues. SPOCK1 often associates with certain complexes binding to proteins like fibronectin and laminin which are important for tissue integrity. Its function can impact cell signaling and tissue remodeling reflecting its diverse biological roles.
Pathways
SPOCK1 plays significant roles in the Wnt signaling pathway and the tissue inhibitors of metalloproteinases (TIMPs) pathway. In these pathways SPOCK1 modulates activities of proteins such as MMP2 and MMP9 which are pivotal for extracellular matrix degradation and remodeling. Its interaction with Wnt proteins highlights SPOCK1's influence in developmental and disease processes requiring precise cellular communication.
SPOCK1 links to cancers and fibrotic diseases. In cancer its overexpression often correlates with tumor progression and metastasis interacting with proteins like integrins to facilitate these processes. In fibrosis altered SPOCK1 levels affect extracellular matrix composition and deposition in conjunction with proteins like collagen contributing to tissue scarring and dysfunction. Understanding SPOCK1's roles provides insight into these conditions' pathology.
