MTERFD1/MTERF3 also known as transcription termination factor 1 or 3 is a member of the MTERF protein family. This protein has a molecular mass of approximately 44 kDa. It localizes primarily in the mitochondria where it functions in regulating mitochondrial transcription. MTERFD1/MTERF3 plays an important role in halting transcription; it binds to specific DNA sequences and impedes the progression of RNA polymerase. This regulation is critical for maintaining mitochondrial homeostasis.
Biological function summary
MTERFD1/MTERF3 regulates mitochondrial gene expression. It forms part of a larger complex involved in the transcriptional control of mitochondrial DNA. This process impacts the synthesis of essential components of the mitochondrial respiratory chain. Through its interaction within this complex MTERFD1/MTERF3 helps maintain normal mitochondrial function which is important for energy production and cellular metabolism.
Pathways
MTERFD1/MTERF3 is an integral part of mitochondrial transcription regulation pathways. It interacts with components of the mitochondrial transcriptional machinery ensuring the orderly expression of mitochondrial genes. Its activity is closely linked with the mitochondrial replication and transcription complex working alongside proteins such as mtDNA-binding protein and TFAM which are important for mitochondrial DNA maintenance and regulation.
MTERFD1/MTERF3 associations are noted in mitochondrial dysfunctions and related syndromes such as MELAS syndrome. Abnormalities in MTERFD1/MTERF3 function can lead to disrupted mitochondrial transcription and compromised respiratory chain function. In these contexts it interacts with proteins like POLG and SUV3 which are implicated in various mitochondrial disorders emphasizing its role in maintaining mitochondrial integrity.
