The Placental Growth Factor or PlGF is an important protein involved in angiogenesis and vasculogenesis. This protein with a molecular mass of around 23 kDa belongs to the vascular endothelial growth factor (VEGF) family. Primarily it expresses in the placenta but you can also find it in the heart thyroid and lungs. PlGF interacts with its receptor VEGFR-1 (Flt-1) playing a significant role in blood vessel formation and repair. This makes it an important focus in the study of vascular biology and pathophysiology.
Biological function summary
PlGF enhances cellular proliferation and migration. As a homodimer it binds to VEGFR-1 mediating angiogenic processes that involve endothelial cells. PlGF's role becomes important in the remodeling of vascular tissues especially under pathological conditions where it supports the growth of new blood vessels. Its expression often increases during tissue ischemia indicating its involvement in tissue recovery and regeneration processes.
Pathways
PlGF functions in the angiogenesis signaling pathway working with proteins such as VEGF-A. This pathway is essential for the development of new blood vessels allowing the supply of nutrients and oxygen to tissues especially during growth or repair. PlGF also plays a role in the inflammatory pathway influencing immune cell activation and infiltration. Through these processes PlGF collaborates with other growth factors and cytokines contributing to a complex network of cellular activities required for maintaining vascular health.
PlGF shows relevance to cancer and cardiovascular diseases. The protein contributes to tumor angiogenesis providing tumors with the blood supply needed for growth and metastasis. PlGF also relates to disorders like atherosclerosis where it influences plaque formation and progression in the arteries. In these contexts PlGF often interacts with other members of the VEGF family modulating the pathological environment. Understanding these interactions offers potential therapeutic targets for managing these conditions better.
