P114RhoGEF also known as ARHGEF18 is a guanine nucleotide exchange factor (GEF) specific for Rho GTPases. It functions by facilitating the exchange of GDP for GTP on RhoA a process important for activating RhoA. p114RhoGEF is approximately 114 kDa in mass. It is expressed in various tissues including vascular endothelial cells and neurons indicating a diverse range of functional roles across different systems.
Biological function summary
This protein plays an important role in the regulation of the actin cytoskeleton dynamics and cell migration. p114RhoGEF associates with other proteins to form complexes that regulate cell shape and motility. It binds with G-proteins and other signaling molecules influencing cellular responses to environmental cues such as changes in cell adhesion or polarity.
Pathways
P114RhoGEF is involved in the Rho signaling pathway which is essential for maintaining cell structure and function. Within this pathway it interacts closely with proteins like ZO-1 and Gα12/13 to control stress fiber formation and focal adhesion. It also features in pathways regulating epithelial cell junctions and endothelial barrier integrity which are critical for tissue homeostasis and vascular function.
P114RhoGEF plays a notable role in cancer progression and cardiovascular diseases. Disruptions in its function can lead to aberrant cell migration and invasion factors linked to tumor metastasis. Additionally changes in its activity might influence endothelial barrier function contributing to vascular leakiness associated with inflammatory conditions. Related proteins such as RhoA and ZO-1 also partake in these pathological processes highlighting the significance of p114RhoGEF in health and disease contexts.
