CPNE3 also known as Copine-3 is a calcium-dependent membrane-binding protein. This protein has a molecular mass of approximately 58 kDa and belongs to the copine family. It consists of two common elements the C2 domain and the A domain known as Von Willebrand factor type A (VWA). CPNE3 expresses in tissues like brain heart and skeletal muscle. These domains enable CPNE3 to associate with cellular membranes in response to calcium signaling implying a role in intracellular signal transduction.
Biological function summary
CPNE3 influences cellular processes such as membrane trafficking signal transduction and cytoskeletal organization. It is not currently known to be a part of any complex but it is thought to mediate interaction between various cellular components through its modular domains. Its role in signal transduction links it to processes like cell movement apoptosis and differentiation. CPNE3 potentially triggers cellular responses by interacting with other proteins and lipids within the cell environment.
Pathways
CPNE3 actively participates in signaling related to calcium influx and integrin pathways. It is associated with the calcium signaling pathway that is significant for various cellular activities like muscle contraction and neurotransmitter release. Additionally CPNE3's interaction within the integrin signaling pathway implicates its role in extracellular matrix adhesion and cellular assembly. Through these pathways CPNE3 interacts with proteins such as integrins and calmodulin influencing cell adhesion and motility.
CPNE3 relates closely to cancer and cardiovascular diseases. Its increased expression in some cancers suggests a connection to tumor progression and metastasis particularly in ovarian cancer and glioblastoma. CPNE3 is connected to proteins such as EGFR and Akt in the context of cancer which are key players in cell proliferation and survival pathways. In cardiovascular diseases its capability to influence muscle function relates to proteins involved in calcium homeostasis impacting conditions like hypertrophy.
