Phosphodiesterase 6C (PDE6C) also known as cone phosphodiesterase is an important enzyme in the phototransduction pathway. It functions mechanically by hydrolyzing cyclic GMP (cGMP) into GMP a critical step in the visual signal transduction process. The PDE6C protein has a molecular mass of approximately 99 kDa. It is chiefly expressed in cone photoreceptor cells of the retina playing a pivotal role in converting light into electrical signals essential for vision.
Biological function summary
PDE6C plays a significant part in the visual process by regulating levels of cGMP in cone cells. It forms a critical component of the phototransduction cascade where it associates with other subunits including alpha beta and gamma subunits to form a functional enzyme complex. This complex adjusts cGMP levels in response to light exposure contributing to the adaptation of photoreceptors to varying light conditions helping modulate visual acuity and color differentiation.
Pathways
PDE6C is deeply embedded in the phototransduction pathway an important cascade for light perception. The hydrolysis of cGMP by PDE6C interacts with the cyclic nucleotide-gated (CNG) ion channels regulating the flow of ions and influencing the membrane potential of cone cells. It shows a relationship with other phototransduction proteins such as guanylate cyclase complementing the regulation of cGMP within the visual signaling pathway.
PDE6C mutations or malfunctions can lead to cone dystrophies and achromatopsia both impacting vision. These conditions result from disrupted cGMP levels in cone cells impairing normal phototransduction. Dysfunctions in PDE6C have indirect associations with GNAT2 another protein critical to phototransduction; this linkage influences the same pathway leading to compromised visual signal processing and associated retinal diseases.
