HOOK3 also known as Hook homolog 3 is a protein with a mass of approximately 90 kDa. This protein functions mechanically as a microtubule-binding protein which plays a role in cytoskeleton organization. In terms of cellular location researchers have identified its expression in various tissues with high presence noted in brain tissue. HOOK3 engages in hetero-oligomeric interactions and contributes to the stabilization of organelles within the cell.
Biological function summary
HOOK3 contributes to cellular functions by facilitating endosomal transport acting as a part of multi-protein complexes. One such complex includes dynein-dynactin motor proteins which aid in intracellular cargo movement along microtubules. HOOK3 anchors vesicles to microtubules and influences trafficking pathways essential for maintaining cellular homeostasis.
Pathways
The role of HOOK3 integrates into intracellular transport and protein sorting pathways. In particular it participates significantly in the endocytic pathway interacting closely with dynein heavy chain (DYNH1) and dynactin for vesicle trafficking to the lysosome. This process ensures appropriate distribution and degradation of proteins and is linked to the maintenance of cell surface receptors.
Researchers have associated HOOK3 with neurodegenerative diseases and certain cancers. In neurodegenerative conditions the improper function of HOOK3 disrupts neuronal cargo transport implicating it in diseases such as Alzheimer's where related proteins like tau are affected. In cancers altered HOOK3 expression can influence cell division potentially involving proteins like p53 in cellular stress responses. Understanding these connections may offer insights into targeted therapeutic strategies.
