ASPM also known by its alternative name "Abnormal Spindle Microtubule Assembly" is a protein that plays an important mechanical role in cell division. It is involved in mitotic spindle formation contributing to chromosome segregation during mitosis. The ASPM protein has a mass of approximately 410 kDa. This protein is expressed notably in neural tissues and regions of high cellular proliferation such as the brain and other rapidly dividing cell populations.
Biological function summary
ASPM supports proper neural development by maintaining the orientation of the mitotic spindle during neurogenesis. It does not function alone but as part of a protein complex involved in regulating mitotic spindle assembly. It associates with other mitotic spindle-associated proteins to ensure precise cell division. Through this it influences cell fate decisions and proliferation within developing tissues.
Pathways
ASPM plays a significant role in the cell cycle particularly during the mitosis phase. It is an essential part of the mitotic spindle checkpoint pathway which ensures accurate chromosome alignment and segregation. ASPM interacts with proteins like CDK1 (Cyclin-dependent kinase 1) to modulate these processes. This involvement secures ordered progression through cell division and prevents aneuploidy by ensuring spindle apparatuses function correctly.
ASPM dysfunction is closely linked to microcephaly where individuals have a significantly reduced brain size. Genetic mutations in the ASPM gene lead to some forms of primary microcephaly. The protein's influence on neurogenesis ties closely to this condition. Additionally ASPM has associations with cancer particularly in tumors where abnormal cell proliferation occurs. Alterations in ASPM expression can coincide with other cancer-related proteins such as TP53 revealing its potential role as a biomarker in tumor development and growth.
