Alpha Galactosidase Activity Assay Kit ab239716 provides a simple, rapid way to monitor total alpha galactosidase (alpha-Gal) activity in a wide variety of biological samples.
How the assay works
In this kit, α-Gal cleaves a synthetic specific substrate releasing a fluorophore, which can be easily quantified (Ex/Em= 360/445 nm). The assay is specific, sensitive and can detect as low as 0.1 μU of α-Galactosidase activity. This kit does not detect beta galactosidase activity.
Alpha Galactosidase assay methods
There are two methods used for alpha galactosidase assays:
The most common method uses 4-Methylumbelliferyl-alpha-D-galactopyranoside, as an alpha galactosidase substrate. Alpha galactosidase cleaves the substrate releasing fluorescent Methylumbelliferone. This is the method used in alpha galactosidase assay kit ab239716.
The alternative, less-popular method uses the substrate 4-nitrophenyl-α D galacto-pyranoside. Similarly, alpha galactosidase cleaves the substrate releasing pNA, which can be detected by absorbance.

Alpha-galactosidase also known as alpha-D-galactosidase or A-galactosidase is an enzyme that catalyzes the hydrolysis of terminal alpha-galactosyl moieties from glycolipids and glycoproteins. It has a molecular mass of approximately 50000 Daltons. This enzyme is expressed in various tissues including the liver heart and kidneys. It functions by efficiently removing alpha-galactose residues from target molecules a process important for maintaining cellular function.
