The target EN1 also known as Engrailed 1 is a homeobox-containing transcription factor involved in developmental processes. EN1 protein weighs approximately 40 kDa and is expressed primarily in the developing brain especially in the midbrain and hindbrain regions during embryonic development. It also appears in the skeletal and nervous systems where it regulates specific gene expression patterns. The protein sometimes referred to as EN1 protein plays a significant role in tissue specification and morphogenesis during early developmental stages.

The Engrailed 1 protein influences pattern formation by controlling the transcription of key developmental genes. Part of several transcriptional complexes it binds to DNA sequences aiding in the specification of positional identities during embryogenesis.
