STIM2 also known as Stromal Interaction Molecule 2 is a protein involved in calcium signaling with a mass of approximately 82 kDa. It is expressed in various tissues including the brain where it functions to regulate calcium entry into cells. STIM2 acts as a sensor that detects decreases in intracellular calcium levels and helps mediate the opening of calcium channels in the plasma membrane.
Biological function summary
The STIM2 protein facilitates calcium homeostasis by working in cooperation with other proteins to form complexes. It plays an essential role in cellular processes by maintaining basal calcium levels. In neuronal cells STIM2 helps regulate synaptic activity and plasticity impacting learning and memory functions. The protein's ability to form complexes and modulate calcium stores is critical for effective cellular signaling.
Pathways
STIM2 is a fundamental component of the store-operated calcium entry (SOCE) pathway which maintains cellular calcium levels. It functions alongside its homolog STIM1 to activate calcium release-activated calcium channels such as ORAI1 facilitating calcium influx into the cell. This regulation is important for the immune responses and muscle function where precise calcium signaling is necessary for optimal cellular performance.
STIM2 has been implicated in neurological disorders such as Alzheimer's disease where altered calcium signaling can contribute to disease progression. STIM2 dysregulation may affect synaptic stability and memory formation leading to cognitive impairment. Additionally the protein associates with disorders involving immune system dysfunction where abnormal SOCE activity plays a role. Its interaction with STIM1 and ORAI1 in these conditions underlines the importance of balanced calcium homeostasis for proper cellular function.
