KLHL35 also known as Kelch-like protein 35 is a protein encoded by the KLHL35 gene in humans. It consists of 615 amino acids leading to a molecular mass of approximately 68 kDa. This protein contains a BACK domain a BTB domain and several Kelch repeats. KLHL35 is expressed in various tissues with notable expression levels in the kidneys and liver. Its structural domains suggest roles in protein-protein interactions and cellular organization.
Biological function summary
KLHL35 functions as part of an E3 ubiquitin ligase complex. Key activities of this complex include tagging specific proteins for degradation via the ubiquitin-proteasome system. By directing the proteolytic pathways KLHL35 plays a part in the regulation of protein turnover and cellular homeostasis. Its presence within this complex indicates a role in maintaining cellular protein quality control influencing various cellular processes.
Pathways
KLHL35 significantly contributes to the ubiquitin-proteasome pathway. It interacts with various proteins by facilitating their ubiquitination and subsequent degradation. Through these interactions KLHL35 influences essential cellular processes linked to protein homeostasis. Another pathway where KLHL35 plays a role is the cell cycle regulation interacting with proteins such as CUL3. These pathways highlight KLHL35's engagement in maintaining cellular functions and protein regulation.
KLHL35 has possible connections to certain kidney disorders due to its expression in renal tissues. Its role in protein degradation suggests that mutations or dysregulation in KLHL35 could affect cellular homeostasis. Additionally aberrant KLHL35 functioning is potentially linked to liver-related conditions. Alterations in the ubiquitination pathways may result in the accumulation or misfolding of proteins that contribute to these ailments possibly involving proteins like p53 which are known for cellular stress response.
