PEX3 also known as Peroxisomal biogenesis factor 3 is a protein involved in peroxisome formation. PEX3 has a known amino acid sequence that gives it a molecular mass of approximately 45 kDa. It is mainly expressed in tissues with high metabolic activity such as liver kidney and heart. As a membrane protein PEX3 is located on the peroxisomal membrane and plays a direct role in the import of proteins into the peroxisome.
Biological function summary
PEX3 is essential for the protein import machinery of peroxisomes. It acts as a receptor on the peroxisomal membrane interacting with other peroxisomal proteins including PEX19. This interaction helps in the assembly of peroxisomal membrane proteins. It is part of a larger protein complex necessary for the formation and maintenance of functional peroxisomes which are organelles responsible for various metabolic functions.
Pathways
PEX3 plays a central role in peroxisome biogenesis. It functions within the peroxisomal protein import pathway an important process for the maintenance of peroxisomes. In this context PEX3 interacts closely with PEX19 which acts as a chaperone and receptor for newly synthesized peroxisomal membrane proteins. Together PEX3 and PEX19 facilitate the import and proper localization of these proteins into the peroxisomal membrane.
Mutations or deficiencies in PEX3 can lead to peroxisome biogenesis disorders (PBDs) such as Zellweger syndrome. This severe genetic disorder affects multiple systems and results from impaired peroxisomal formation and function. PEX1 another protein involved in peroxisome biogenesis also relates to these disorders emphasizing the importance of coordinated interactions between these proteins for proper cellular function.
