FBXO2 also known as F-box protein 2 is part of the F-box protein family. This protein plays a mechanical role in recognizing specific substrates for ubiquitination a process important for protein degradation. FBXO2 weighs around 32 kDa and mainly expresses in the brain where it regulates protein levels. This localization suggests its significance in neural protein management.
Biological function summary
FBXO2 participates in forming the SCF (SKP1-CUL1-F-box protein) complex a multi-protein E3 ubiquitin ligase complex. It targets glycosylated proteins for degradation which helps control glycoprotein homeostasis in cells. FBXO2 binds to N-linked glycoproteins indicating its specificity towards glycan recognition and processing. This function makes it important for maintaining cellular protein balance.
Pathways
FBXO2 is involved in the ubiquitin-proteasome pathway which is essential for protein turnover and quality control. It also plays a role in the endoplasmic reticulum-associated degradation (ERAD) pathway which manages misfolded proteins. FBXO2 collaborates with proteins such as β-transducin repeat-containing protein (β-TrCP) within these pathways showing its interaction in various cellular mechanisms.
FBXO2 has connections to neurodegenerative disorders and cancer. Aberrations in FBXO2 function are linked to Alzheimer's disease due to its role in protein degradation in neurons. In cancer FBXO2 alteration affects glycoprotein regulation potentially leading to tumor progression and metastasis. The protein relates to other F-box proteins which also influence cellular stability and disease progression.
