SH3GL2 also known as Endophilin A1 is a protein that plays an important role in membrane dynamics during endocytosis. It has a molecular weight of approximately 41 kDa. Mechanically SH3GL2 binds to lipids and helps in membrane curvature due to its BAR domain. This protein is highly expressed in neuronal tissues where it participates in synaptic vesicle endocytosis. It interacts with other proteins via its SH3 domain facilitating the assembly of protein complexes involved in these cellular processes.
Biological function summary
SH3GL2 is involved in synaptic vesicle recycling an important process for neurocommunication. It is part of a complex that includes proteins such as dynamin and synaptojanin. It ensures the rapid turnover of synaptic vesicles by mediating their endocytosis and recycling back to the synaptic terminal. This function helps in maintaining efficient synaptic transmission essential for neuronal communication and plasticity.
Pathways
SH3GL2 is involved in the clathrin-mediated endocytosis pathway. This pathway is critical for the retrieval of synaptic vesicles in neurons. SH3GL2 works closely with clathrin and dynamin to facilitate the budding and scission of vesicles from the plasma membrane. In addition SH3GL2 also interacts within the synaptic vesicle cycle ensuring the availability of vesicles for neurotransmitter release.
SH3GL2 has been linked to neurodegenerative diseases such as Parkinson's disease. Its interaction with proteins like alpha-synuclein suggests its role in the pathogenesis of this disorder. Altered SH3GL2 function may disrupt normal synaptic vesicle dynamics contributing to neuronal dysfunction. Furthermore aberrant SH3GL2 expression or function may also be involved in schizoaffective disorders where its interaction with synaptojanin plays a role in synaptic dysfunction.
