Prostaglandin E Synthase (MPGES-1) also known as prostin E is a membrane-associated protein that catalyzes the conversion of prostaglandin H2 to prostaglandin E2. It has a molecular mass of about 16 kDa. MPGES-1 is widely expressed in various tissues including the kidneys lungs and heart. The expression of MPGES-1 increases in response to pro-inflammatory signals highlighting its adaptive response during inflammation.
Biological function summary
Prostaglandin E Synthase plays an important role in mediating inflammatory responses and is part of the terminal enzyme complex in the prostaglandin synthesis pathway. Its activity contributes to the elevated production of prostaglandin E2 (PGE2) a bioactive lipid that regulates inflammation fever and pain. MPGES-1 functions in concert with Cyclooxygenase-2 (COX-2) as part of the coordinated induction during inflammatory stimuli.
Pathways
The MPGES-1 enzyme is central to the arachidonic acid cascade specifically in the cyclooxygenase pathway. This pathway leads to the production of prostaglandins which are involved in inflammatory and immune responses. Prostaglandin E2 produced by MPGES-1 is related to proteins such as COX-2 and cytosolic phospholipase A2 (cPLA2) which precede it in the biosynthesis pathway and act collaboratively in the metabolism of arachidonic acid to prostanoids.
MPGES-1 is implicated particularly in conditions like rheumatoid arthritis and cancer. Elevated levels of prostaglandin E2 produced by the concerted action of MPGES-1 have been observed in inflamed joints of arthritis patients correlating with COX-2 expression. In cancer upregulation of MPGES-1 contributes to tumorigenesis through promoting angiogenesis and proliferation. Its interaction with COX-2 enhances oncogenic processes signifying its potential as a therapeutic target.
