Transmembrane Emp24 protein transport domain containing 2 also known as TMED2 is a protein that plays an important role in the transport of proteins between the endoplasmic reticulum (ER) and the Golgi apparatus. TMED2 weighs approximately 23 kDa. Research indicates TMED2 as highly expressed in various tissues showing a significant presence in the liver and kidney. It acts as part of the p24 family of proteins which are involved in cargo trafficking within intracellular membranes.
Biological function summary
TMED2 facilitates the sorting and transportation of proteins by forming a coat around vesicles enabling their movement between cellular compartments. It is an essential member of the p24 protein complex which functions as a carrier in the early secretory pathway. This complex supports the maintenance of cellular protein balance and ensures proteins reach their proper destinations efficiently.
Pathways
TMED2 contributes to the vesicular trafficking pathway which controls the flow of proteins within the cell. It participates in the secretory pathway closely interacting with COPI and COPII proteins that form vesicle coats important for cargo selection and vesicle budding. TMED2 interacts particularly with proteins like TMED10 and TMED9 forming a network that underpins its role in vesical transport from the ER to the Golgi.
TMED2 has connections to certain metabolic disorders and cancer. Improper function or expression of TMED2 can lead to defects in protein trafficking which can disrupt cellular homeostasis and contribute to conditions like congenital disorders of glycosylation. Moreover alterations in TMED2 levels have been observed in some cancer types such as colorectal cancer.
