RIOK1 also known as RIO1 is an atypical serine/threonine kinase with a molecular mass of approximately 67 kDa. It plays a role in processing small ribosomal subunit biogenesis. The protein is expressed in various tissues including liver and heart. Unlike typical kinases RIOK1 does not rely on the classical ATP-dependent mechanism for its kinase activity. It participates in the phosphorylation of some proteins which is necessary for ribosome maturation.

The kinase RIOK1/RIO1 is part of the ribosome biogenesis pathway which is essential for protein synthesis. It forms part of a larger dynamic complex that assists in the maturation and transport of ribosomal components from the nucleus to the cytoplasm.
