Human Granulysin ELISA Kit (ab256402) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Granulysin protein in cell culture supernatant, cit plasma, edta plasma, and serum. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human Granulysin with 8.22 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Granulysin (GNLY) is a 15.5 kD protein is a member of the saposin-like protein (SAPLIP) family. It is mapped to 2p11.2. Granulysin functions to create holes in the target cell membrane and destroy it. It is a microbiocidal via induction of apoptosis. The gene is expressed in cytolytic granules with perforin, a pore forming protein as well as granzymes that are also involved in cytolysis. In addition to it. As an antimicrobial, Granulysin’s activities include killing microbes that cause tuberculosis and malaria. It has been reported to be involved in tumor destruction. A series of peptides generated from the amino acid sequence of Granulysin are potential antibiotics. Granulysin shares 76% and 26% sequence homology with monkey and mouse, respectively.

Granulysin also known as GNLY is a cytolytic protein with a mass of approximately 9 kDa present in the granules of cytotoxic T lymphocytes and natural killer cells. This protein expressed in activated human immune cells plays a role in immune defense by creating pores in the target cell membranes which enables other cytotoxic molecules to enter and induce cell death. Researchers often utilize Granulysin as a GNLY marker to assess immune cell activity within the body.
Biological function summary
Granulysin contributes to the body's defense against bacterial fungal and parasitic infections. It acts independently but can also collaborate with other immune components to enhance its effects. Granulysin disrupts microbial membranes owing to its affinity to negatively charged lipids leading to pathogen elimination. This mechanism plays a critical role in the human body's broader immune response.
Pathways
Granulysin operates significantly within the immune response pathway by working in concert with perforin another cytolytic protein. Together they form a potent immune mechanism aimed at eliminating infected or malignant cells. Furthermore the interferon-gamma pathway closely influences Granulysin suggesting a connection to innate and adaptive immune responses.
Granulysin shows association with various conditions most notably tuberculosis and leprosy. In these diseases the GNLY protein levels reflect the body's immune activity against the pathogens.
