The protein ORC2L also known as Origin Recognition Complex Subunit 2 plays a role in DNA replication initiation. It has a mass of approximately 62 kDa. This protein is widely expressed in various tissues with notable abundance in proliferative cells. Within the cell ORC2L localizes to the nucleus and assists in defining the replication origins to begin DNA synthesis. It binds to DNA sequences contributing to the organization of chromatin structure to facilitate replication.
Biological function summary
This protein acts as part of the Origin Recognition Complex (ORC) which is essential for DNA replication initiation. ORC2L works with other ORC subunits to form a platform for the subsequent recruitment of additional factors necessary for DNA helicase loading. These sequential events prepare for unwinding the DNA double helix allowing DNA polymerases access during the S phase of the cell cycle. The integrity and function of ORC2L ensure accurate DNA replication and help maintain genomic stability.
Pathways
ORC2L participates in the DNA replication pathway and the cell cycle regulation pathway. In the DNA replication pathway ORC2L cooperates with proteins like CDC6 and CDT1 which further stabilize the pre-replication complex. ORC2L's activity manages the licensing of replication origins ensuring proper timing of DNA synthesis. Its role within the cell cycle regulation pathway aids in coordinating the replication events with cell cycle progression impacting the transition from the G1 to the S phase.
Research links ORC2L to certain cancers and Meier-Gorlin Syndrome. In the context of cancer alterations in the expression or function of ORC2L can lead to genomic instability and unregulated cell proliferation which are hallmarks of cancerous transformations. Meier-Gorlin Syndrome a disorder characterized by developmental abnormalities results from mutations affecting ORC2L or other related proteins within the origin recognition complex such as ORC1 and CDT1. These mutations disrupt normal pre-replication complex function leading to impaired cell division and growth abnormalities.
