ECE2 also known as Endothelin-Converting Enzyme 2 is a metalloprotease with a molecular mass of approximately 85 kDa. It plays a role in converting inactive big endothelins into active endothelins which are potent vasoactive peptides. You find ECE2 expressed in various tissues including the brain heart and lungs. The protein localizes mainly to the cell membrane and endosomal compartments where it performs its enzymatic functions.
Biological function summary
ECE2 has a significant role in the regulation of vascular tone and blood pressure. It is an integral part of the endothelin system not forming a complex but working along with endothelin receptors to modulate vasoconstriction and vasodilation. Its activity helps maintain cardiovascular homeostasis. The enzyme processes peptides that influence neuronal signaling suggesting a role in neural function as well.
Pathways
ECE2 features prominently in the endothelin signaling pathway and the broader cardiovascular regulatory pathway. It processes the precursor proteins big endothelins which then activate endothelin receptors (EDNRA EDNRB). These receptors further engage with G-proteins to promote downstream signaling that regulates vascular constriction and blood pressure. ECE2 activity is important for ensuring the balance of these signaling cascades particularly in cardiovascular and neural contexts.
ECE2 plays a part in the pathophysiology of hypertension and Alzheimer's disease. Altered ECE2 activity influences endothelin levels contributing to blood pressure dysregulation and hypertension. In Alzheimer's disease ECE2 works with proteins like beta-amyloid peptide where dysregulation may lead to neurotoxicity and plaque formation. These connections make ECE2 a potential therapeutic target in disease intervention.
