UBE2D4 also known as UBC4/5 homolog is an enzyme with a molecular weight of approximately 16 kDa. This protein functions as a ubiquitin-conjugating enzyme that plays a role in the ubiquitination process helping to transfer ubiquitin from an E1 activating enzyme to an E3 ligase substrate protein. UBE2D4 is expressed in various tissues throughout the body including high expression in skeletal muscle and liver. Its mechanical role is critical in tagging proteins for degradation via the proteasome.
Biological function summary
The ubiquitin-conjugating enzyme plays a role in protein homeostasis by facilitating the ubiquitin-proteasome system a complex that includes E1 E2 and E3 enzymes. UBE2D4 acts in concert with specific E3 ligases to regulate cell cycle progression and transcriptional regulation. This process ensures the removal of misfolded or damaged proteins thereby maintaining protein quality within cells. Its involvement in diverse cellular pathways highlights the enzyme's importance in controlling cellular activities.
Pathways
UBE2D4 operates in the ubiquitin-mediated proteolysis and cell cycle regulatory pathways. These pathways control protein turnover and degradation ensuring cellular equilibrium. UBE2D4 collaborates with E3 ligases like MDM2 within these pathways influencing proteins like p53 which is important for cell cycle control and apoptosis. The interconnectedness with MDM2 emphasizes UBE2D4's role in key cellular events.
Alterations or malfunctions in UBE2D4 activity have linked it to cancer and neurodegenerative diseases. Misregulation of p53 related proteins via UBE2D4 alterations may promote unrestrained cell proliferation seen in cancers. Furthermore abnormal ubiquitination processes associating with pathways including UBE2D4 contribute to the accumulation of protein aggregates in neurodegenerative disorders including Alzheimer's disease. The relationship with MDM2 and p53 proteins highlights UBE2D4's significant role in disease pathogenesis.
