SELH also known as Selenoprotein H is a selenocysteine-containing protein with a molecular mass of approximately 15 kDa. It functions in redox regulation and protection against oxidative stress by facilitating antioxidant defense mechanisms. SELH is mainly expressed in the brain and to a lesser extent in other tissues. It belongs to the selenoprotein family which includes proteins containing the rare amino acid selenocysteine responsible for its unique enzymatic capabilities.
Biological function summary
Selenoprotein H contributes to protecting cells from oxidative damage. It plays a role in regenerating antioxidant molecules and maintaining cellular redox balance. SELH is not known to be part of a larger protein complex but works in concert with other antioxidant molecules to enhance cellular resistance to oxidative stress. It also involves gene regulation particularly through modulating transcription factors that respond to oxidative signals.
Pathways
Selenoprotein H engages significantly in the cellular antioxidant pathways and redox homeostasis. It interacts with enzymes like glutathione peroxidase and thioredoxin reductases which are essential in countering oxidative stress. These interactions help maintain signal transduction pathways which depend on redox-sensitive components affecting cellular metabolism and survival. SELH along with these proteins ensures proper cellular function by modulating oxidative status and impacting pathways related to energy metabolism and apoptosis.
Selenoprotein H relates notably to neurodegenerative diseases and certain cancers. SELH's role in protecting neurons from oxidative stress connects it to conditions like Alzheimer's disease where oxidative stress contributes to neuronal degradation. Additionally it is involved with the tumor suppressor protein p53 which responds to oxidative stress in cancer development. SELH's antioxidant activity can influence these pathways impacting disease progression and therapeutic responses.
