The eukaryotic translation initiation factor 4H commonly known as eIF4H is an essential protein participating in the initiation phase of protein synthesis. It holds a mass of approximately 27 kDa. eIF4H is expressed in various tissues including the brain liver and muscle. The protein promotes the unwinding of RNA secondary structures aiding ribosome attachment to the mRNA. This function aligns closely with its alternate name eukaryotic translation initiation factor 4H.
Biological function summary
EIF4H enhances the helicase activity of eIF4A an RNA helicase vital for translation initiation. It forms part of the translation initiation complex. The complex brings together eIF4E eIF4G and eIF4A enhancing mRNA recognition and processing. eIF4H stabilizes the eIF4A-mRNA interaction reducing the likelihood of premature dissociation and permitting efficient scanning for the start codon.

EIF4H plays a significant role in the mTOR (mechanistic target of rapamycin) signaling pathway which regulates cell growth and proliferation. This pathway influences protein synthesis by controlling the availability and activity of initiation factors like eIF4A and eIF4E. mTOR signaling overlaps with the MAPK (mitogen-activated protein kinase) pathway with eIF4H indirectly interacting with RAF proteins which regulate cell cycle progression and differentiation.
Researchers link eIF4H to neurological disorders like Autism Spectrum Disorder and certain cancers such as glioblastoma. Alterations in the expression or function of eIF4H may impact the mTOR pathway influencing tumor progression and neural development. These dysfunctional roles also involve eIF4E as both proteins contribute to abnormal translation initiation and gene expression in these conditions.
