Mouse H60a ELISA Kit is designed for the quantitative determination of H60a in cell culture supernatants, plasma and serum samples.

Mouse H60a concentration is low in normal serum/plasma, and may not be detectable in this assay.
This assay employs an antibody specific for mouse H60a coated on a 96-well plate. Standards and samples are pipetted into the wells and H60a present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-mouse H60a antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of H60a bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

H60 also known as H60a is a murine non-classical MHC class Ib molecule with a mass of approximately 34 kDa. This protein plays a role as a surface ligand for the activating receptor NKG2D which is part of the immune system. H60a is mainly expressed on the surface of activated cells such as T cells and also on transformed cells and stressed cells. Its expression varies in different tissue types including the spleen and the small intestine reflecting its important role in immune surveillance.
Biological function summary
H60 acts as an activating ligand for NKG2D receptors on natural killer (NK) cells and some T cells. When H60 binds to the NKG2D receptor it induces an immune response which can lead to the destruction of the target cells presenting H60. H60 is not typically part of any larger complex but functions through its interaction with NKG2D promoting cellular immune response against infected or transformed cells.
Pathways
H60 is influential in immune signaling pathways especially those related to the cytotoxic response. It plays a part in the NKG2D receptor-ligand interaction pathway interacting with other proteins like the RAET1 family which also function as ligands for NKG2D enhancing cellular cytotoxicity against cells displaying stress-induced antigens. This mechanism is key to the regulation of immune responses and maintaining immune equilibrium.
H60's expression has links to cancer and viral infections. In tumors elevated H60 expression can mark malignant cells for NK cell-mediated destruction an important defense mechanism against cancer progression. Additionally in similar ways during viral infections infected cells may express H60 therefore alerting the immune system and promoting anti-viral responses. Through these roles H60's interaction with NKG2D highlights its importance in disease contexts where immune surveillance is critical for disease management.
