PPP2R1A also referred to as PP2A alpha is a regulatory subunit of the protein phosphatase 2A (PP2A) complex. The protein has a mass of approximately 65 kDa. It functions mechanically as a scaffold subunit within the PP2A holoenzyme serving to coordinate the interaction between catalytic and regulatory subunits of the complex. This protein is widely expressed across various tissues which reflects its role in numerous cellular processes.
Biological function summary
The activity of PPP2R1A is essential for the dephosphorylation of serine/threonine residues on proteins allowing significant regulation of various cellular functions. The protein is an important part of the PP2A complex a heterotrimer that consists of structural (A) catalytic (C) and regulatory (B) subunits. This complex contributes to the control of cell growth and division by maintaining appropriate levels of phosphorylation within the cell impacting various signaling pathways.
Pathways
The functional activity of PPP2R1A integrates into critical pathways like the MAPK signaling pathway and cell cycle regulation. It modulates the activities of kinases and other proteins such as AKT and MEK which are critical to these pathways and are important in processes like cell proliferation and apoptosis. PPP2R1A's regulatory role ensures proper signal transduction within these pathways for maintaining cellular homeostasis.
The malfunction or mutation of PPP2R1A has been linked to cancers such as lung and colorectal cancer. Aberrant signaling within the pathways it regulates can lead to unregulated cell growth and contribute to tumorigenesis. Furthermore the protein's dysfunction can be associated with disorders connected to AKT signaling dysregulation reinforcing the importance of PPP2R1A's role in maintaining cellular structure and function.
