The protein Inositol (13456)-pentakisphosphate 2-kinase commonly known as IPPK or IPK1 plays an important role in the phosphorylation of inositol phosphates. IPPK catalyzes the conversion of inositol pentakisphosphate (IP5) to inositol hexakisphosphate (IP6). This protein has a molecular mass of around 44 kDa. It is prominently expressed in the brain kidney and pancreas suggesting a broad importance across different tissues.
Biological function summary
IPPK contributes to the regulation of inositol phosphate metabolism. It does not act in isolation but as part of a network involving inositol polyphosphate kinases and phosphatases that help maintain cellular processes such as signal transduction and ion homeostasis. Although IPPK doesn't form part of a larger complex its activity impacts various cell signaling pathways by altering the levels of its substrate and product.
Pathways
IPPK participates significantly in the phosphoinositide signaling pathway impacting processes such as cellular growth survival and apoptosis. This pathway also involves PI3Ks and PTEN which are critical in signal transduction. Moreover IPPK indirectly influences the synthesis of IP7 regulated by inositol hexakisphosphate kinases integrating its function into broader metabolic pathways.
IPPK has associations with pathologies like cancer and metabolic disorders. Abnormal activity or expression levels of IPPK can alter cellular signaling pathways which might lead to tumorigenesis or metabolic syndrome. It closely interacts with proteins such as mTOR highlighting its role in cancer pathways and PI3K/AKT pathway dysregulation common in metabolic disorders.
