The GPCR GPR64 also known as ADGRG2 is a member of the adhesion G protein-coupled receptor family. It is characterized by large extracellular domains facilitating cell adhesion and signaling. This receptor has a mass of approximately 98 kDa. GPR64 is mainly expressed in the epididymis and plays an important role in reproductive tissues. Moreover it displays a basal activity that implicates several signaling pathways.
Biological function summary
GPCR GPR64 functions as a mediator of reproductive tract development. It is not currently known to be part of any larger complex. However its signaling contributes to the modulation of fluid reabsorption in the epididymis affecting sperm maturation and storage. Beyond the reproductive system some studies suggest roles in other tissues like osteoblasts and kidney cells.
Pathways
Several pieces of research have tied GPCR GPR64 to the cAMP signaling pathway influencing the regulation of epithelial sodium channels. This impacts ion transport and homeostasis within the reproductive tissues. Additionally other proteins such as epithelial sodium channel (ENaC) have shown interactions with GPR64 affecting fluid dynamics. Further research is exploring potential connections to the Wnt signaling pathway due to similar functional roles.
GPCR GPR64 has been linked to disorders affecting fertility such as reduced sperm motility leading to male infertility. Also abnormal expression patterns have been observed in some cancer types like prostate cancer. The interaction with ENaC makes it particularly interesting in understanding how dysregulation might impact these conditions on a cellular level providing insights for potential therapeutic targets.
