Free Amino Nitrogen Assay Kit (ab272523) measures alpha amino acids, ammonia, and end group amino nitrogens.
Key facts
Detection method:Colorimetric,
Sample types:Food samples,
Assay type:Quantitative,
Range:0.2 - 10 mM,
Assay Platform:Microplate

Product details:
Free Amino Nitrogen Assay Kit (ab272523) measures alpha amino acids, ammonia, and end group amino nitrogens. The ninhydrin based reaction is a superior method for determining only alpha amino acids and ammonia compared to the traditional Kjeldahl, which measures nitrogen from all sources. Only requiring low sample volumes, the stable ninhydrin reagent provides a simple and accurate method for determining Free Amino Nitrogen concentrations.
Fast and sensitive.
Linear detection range (5 μL sample): 0.2 to 10 mM for 10 min reaction.
Convenient and high-throughput.
Homogeneous "mix-incubatemeasure" type assay. Can be readily automated to process thousands of samples per day.

Properties and Storage Information:

Amino nitrogen consists of elemental nitrogen atoms present in amino groups such as NH2 found in amino acids and proteins. It plays a central role in protein structure and function. Alternative names for related compounds include "ninhydrin" which is often used in testing amino acids. Chemical assays like the "ninhydrin test" or "amino assay" help to quantify amino nitrogen by detecting free amino groups. These methods are essential for analyzing proteins expressed throughout the body as amino nitrogen is ubiquitous in all protein-containing tissues.
Biological function summary
Amino nitrogen's presence in proteins and amino acids makes it fundamental to life processes. Amino nitrogen serves as key building block in proteins which are abundant in both structural components and enzymes. Proteins with amino nitrogen participate in forming complex macromolecular structures functioning as catalysts in biochemical reactions. Its role in nitrogen metabolism impacts synthesis and degradation of biological molecules within the cell.
Pathways
Amino nitrogen is integral to both the urea cycle and the nitrogen assimilation pathway. It engages with enzymes to assist in the conversion of ammonia to urea in the liver. The nitrogen assimilation pathway incorporates amino nitrogen for biosynthesis of amino acids aiding processes such as glutamine and glutamate synthesis. These pathways link to proteins like aspartate transcarbamoylase reflecting its broad involvement in metabolic cycles.
Abnormal amino nitrogen metabolism has connections to disorders like hepatic encephalopathy and phenylketonuria. In hepatic encephalopathy improper handling of amino nitrogen leads to toxic accumulation of ammonia due to liver dysfunction implicating proteins such as glutamine synthetase. Phenylketonuria results from faulty metabolism of phenylalanine an amino acid again involving amino nitrogen. These conditions highlight amino nitrogen's significance in disease mechanisms.
