PKC (Protein Kinase C) is a family of serine/threonine kinases involved in a variety of cellular processes. Common isoforms include PKC alpha delta zeta eta theta and iota each with distinct functions. These proteins such as PKC alpha at approximately 80 kDa are expressed across multiple tissues. PKC proteins play a critical role in signal transduction acting through phosphorylation events that regulate diverse cellular functions. The related kinases JAK1 JAK3 and ABL1 further contribute to cellular communication while proteins such as Fyn and RSK4 partake in linking upstream signals to downstream targets.
Biological function summary
PKC isoforms influence processes such as cell proliferation differentiation and apoptosis. They function both independently and as part of larger protein complexes. Specifically CDK2 FAK and PDK1 often interact as part of the pathways regulating these processes. Additionally VEGF Receptor 2 and 3 play significant roles in vascular development and repair while Cytochrome P450 3A4/CYP3A4 is involved with drug metabolism. Other targets like GSK3 beta and MAP4K6 participate in cellular responses to external stimuli.
Pathways
PKC signaling integrates with the MAPK and PI3K-Akt pathways which are important in cell survival and proliferation. The PKC pathway modulates interactions with proteins such as RACK1 which acts as an anchoring protein and NELL1 implicated in osteogenesis. Multiple PKC isoforms influence these pathways allowing for diverse cellular outcomes based on specific signals. GSK3 alpha despite being similar to its beta isoform may interact differently within these pathways highlighting the complexity of PKC signaling.
Altered PKC signaling associates with cancer and cardiovascular diseases. In many cancers aberrant PKC activity contributes to unchecked cell growth and survival. The PKC zeta isoform in particular may influence tumorigenesis often in conjunction with proteins like MAP4K4/NIK and PIM1. VEGF Receptor 2 deregulation is closely linked to angiogenesis disorders commonly found in cancer progression as well. These molecular connections showcase the importance of PKC and related proteins in understanding and potentially targeting disease pathways.
