Cysteine-Rich with EGF-Like Domains 2 (CRELD2) is a protein encoded by the CRELD2 gene also known as Cysteine-Rich with EGF-Like Domains Protein 2. The protein consists of 362 amino acids and has a molecular mass of approximately 39 kDa. CRELD2 expresses mainly in the endoplasmic reticulum and can also be found in many tissues with notable levels in neural tissues and cardiac muscle. This protein functions as a chaperone influencing the folding and stability of other proteins within the cell.
Biological function summary
CRELD2 plays a significant role in the endoplasmic reticulum stress response a process important for maintaining cellular homeostasis. The protein modulates cellular stress pathways and associates with several other chaperone proteins assisting in the proper folding of newly synthesized proteins. It does not associate with a particular protein complex but functions independently to ensure efficient protein synthesis and folding under stress conditions.
Pathways
CRELD2 integrates primarily with the unfolded protein response (UPR) pathway activated during ER stress to restore normal function by halting protein translation and facilitating protein folding. In this pathway CRELD2 interacts with key proteins such as ATF6 and IRE1. These interactions enhance its role in correcting misfolded proteins and prevent the accumulation of toxic protein aggregates within cells.
CRELD2 links to several conditions especially those related to disrupted protein homeostasis like Alzheimer's disease and cardiac abnormalities. Disruptions in CRELD2 expression can impact protein folding and stress responses which contributes to the pathogenesis of these diseases. It also connects with proteins such as amyloid precursor protein (APP) and cardiac myosin emphasizing its role in neuronal and cardiac health.
