Fatty acid-binding protein 6 (FABP6) also known as ileal bile acid binding protein (IBABP) plays a significant role in binding bile acids. This protein weighs approximately 14 kDa and is prominently expressed in the ileum of the small intestine. It functions primarily within the cytoplasm ensuring the transportation of bile acids to specific locations within cells. By doing this FABP6 facilitates the proper metabolism and absorption of these acids which are critical components for the digestion and absorption of dietary fats.
Biological function summary
The protein plays a critical role in the enterohepatic circulation of bile acids which it binds with high affinity. FABP6 operates within a network of ligands and ions aiding in the homeostasis of bile acids. While it does not directly associate with larger complexes its action contributes to maintaining a balance of cholesterol and fats in the body. It also integrates with other fatty acid-binding proteins like FABP1 and FABP3 which engage in similar transport functions in different tissues.
Pathways
FABP6 functions prominently within the bile acid signaling and lipid metabolism pathways. It interacts closely with proteins such as FXR (Farnesoid X receptor) that senses bile acid concentrations and regulates their synthesis and transport. Additionally FABP6 has relationship with the bile acid-CoA:amino acid N-acyltransferase (BAAT) which is involved in bile acid conjugation. These connections underpin its key role in lipid digestion and cholesterol homeostasis.
FABP6 has associations with cholestasis and Crohn’s disease. In cholestasis impaired bile flow can arise from the dysfunction of bile acid transport to which FABP6 is linked. Furthermore alterations in the protein’s expression or function contribute to Crohn’s disease particularly in the ileal region where it heavily expresses. Mutations and malfunction of related proteins such as FXR and BAAT can complement or exacerbate these issues solidifying the importance of FABP6 in maintaining gut and liver health.
