The SARS-CoV-2 envelope protein also known as E protein plays an important role in viral assembly and release. It is a small structural protein with a mass of approximately 8.4 kDa and is expressed on the surface of the coronavirus. E protein is an integral membrane protein that localizes to the endoplasmic reticulum-Golgi intermediate compartment (ERGIC) where it assists in the assembly of the virus envelope. It contains multiple transmembrane domains that enable it to interact with other viral components inside the host cell.
Biological function summary
The SARS-CoV-2 E protein facilitates the virus budding process and contributes to the pathogenicity of the virus. It is a component of the virus envelope which maintains the structural integrity of the virion. E protein is also involved in ion channel activity acting in the formation of viroporins that modulate host cell environments. This ion channel activity aids in virus-host cell interactions and can impact cell signaling pathways advancing the virus's pathology.
Pathways
The SARS-CoV-2 E protein engages in critical processes of viral assembly and pathogenesis. It plays roles in the viral replication pathway and is implicated in the formation of virus-like particles (VLPs). The E protein interacts with other viral proteins such as the membrane protein (M) and nucleocapsid protein (N) coordinating the assembly and packaging of the virion. These interactions are essential for the completion of the viral life cycle within the host cell influencing virus infectivity and stability.
SARS-CoV-2 E protein is directly associated with COVID-19 the disease caused by the coronavirus. The protein contributes to the virulence of the virus and can influence the clinical manifestations of COVID-19. Researchers have explored its links to acute respiratory distress syndrome (ARDS) as it can affect immune responses and inflammation in the lungs. The protein's interaction with the spike protein (S) is critical in understanding the disease's progression as these proteins collectively enable the virus to evade the host immune system and facilitate successful infection.
